T4 DNA Polymerase

T4 DNA Polymerase is a DNA-dependent DNA polymerase which catalyzes the 5¡¯¡æ3¡¯ synthesis of DNA and requires template and primer.  Additionally, the enzyme has a strong 3¡¯¡æ5¡¯ exonuclease activity.
Source:
Purified from a strain of E. coli that carries a T4 DNA polymerase expression plasmid.
Applications:
  • Fill in of 5¡¯ overhanging ends to create blunt-ends (1).
  • Exonuclease removal of 3¡¯ overhanging to create blunt-ends (1).
  • in vitro mutagenesis (2)
Heat inactivation: 70¡ÆC for 15 minutes.
Storage:
Enzyme and buffer should be stored at -20¡ÆC. Enzyme is stable for 12 months if handled properly.
Additional Information:
  • T4 DNA Polymerase is active in many buffers and can often be used directly in a restriction digest reaction.
  • T4 DNA Polymerase exhibits no strand displacement activity.
